چکیده

Introduction Exposure of normal human fibroblasts to ionizing radiation may cause a delayed instability of the genome in the descendants of the initially irradiated cells [1, 2]. This genomic instability appears as a de novo formation of structural chromosomal aberrations and is considered as a step towards malignant transformation and induced carcinogenesis [3]. The relevance of effects, related to genomic instability, observed in different experimental setups, cell strains and the dependence on radiation quality are still under debate [4]. In recent (or previous) experiments we have shown the occurrence of genomic instability in the progeny of mixed cell populations of fibroblasts initially irradiated with high doses of X-rays [2]. To circumvent a selective pressure and population dynamics on the yield and types of chromosomal aberrations in bulk cultures and to investigate the fate of single cells, colonies formed by single cells after irradiation with X-rays were isolated and analysed for cytogenetic changes in the present study.

Chromosomal instability in haemopoietic cells of the foetus, mother and offspring after in utero irradiation of the CBA/Ca mouse.

PURPOSE The present study was conducted to test the susceptibility of the mouse foetus to transmit chromosomal instability to the haemopoietic stem cells of offspring after in utero X-or plutonium-239-irradiation. MATERIALS AND METHODS Pregnant CBA/Ca-mice were injected with 80 kBq/kg 239Pu or X-irradiated with 1 Gy X-rays on days 13 or 14 of gestation. CFU-A cultures were grown from haemopoi...

Induction of cancer stem-like cells in A549 cells after exposure to carbon ions and X-rays

Background: Cancer stem-like cells (CSCs) play a crucial role in the initiation, progression, and recurrence of cancer. Evidence indicates that the high linear energy transfer (LET) carbon ion beam is more effective against CSCs than the conventional X-ray beam. Carbon ion radiotherapy is considered as a promising cancer strategy, however, information about whether, or not, new CSCs are induced...

Genetic instability and telomere shortening in normal human fibroblasts after irradiation with X-rays

Introduction Former experiments have shown an increased number of non-transmissible chromosomal aberrations in the progeny of normal human fibroblasts after irradiation with Xrays [1, 2]. These de novo formed non-transmissible aberrations are an indicator of genomic instability which is considered to be one important step towards carcinogenesis. In parallel experiments we observed accelerated s...

Lethal sectoring, genomic instability, and delayed division delay in HeLa S3 cells surviving alpha- or X-irradiation.

Lethal sectoring (LS) is the process for survival in which lethal damage remaining in irradiated cells is eliminated as lethal sector (offspring without reproductive integrity). This process occurs through the postirradiation 1st to 4th divisions with the accompanying appearance of a clonogenic progenitor (clonogen) (clonogenic sectoring: CS). The features of LS or CS and genomic instability (G...

Genotype-dependent induction of transmissible chromosomal instability by gamma-radiation and the benzene metabolite hydroquinone.

Although it is well established that ionizing radiation and benzene are epidemiologically linked to acute myeloid leukemia (AML), the underlying mechanisms are not understood. We have shown that gamma-radiation can induce a persisting genomic instability in the clonal descendants of hemopoietic stem cells manifested as a high frequency of nonclonal chromosome and chromatid aberrations. A striki...
